HIV-1 Integrase Enzyme

...catalyzes DNA integration of viral DNA into the host-cell genome.

Human immunodeficiency virus type 1 (HIV-1), like all retroviruses, depends upon the integration of a DNA copy of its viral genome into host cell chromosomes as part of its infection cycle. The HIV-1 integrase (IN) enzyme mediates this insertion of viral DNA into the human genome and it is one of three virally-encoded enzymes required for virus replication. Because the proviral integration event is essential for HIV-1 replication, the HIV-1 IN enzyme remains a promising target for antiviral drug development to treat patients with AIDS.

 

 This purified HIV-1 Integrase prepartion is enzymatically active in DNA strand-transfer assays.

HIV-1 Integrase is an affinity-purified enzyme that catalyzes the insertion of proviral DNA sequences into target DNA. This enzyme contains the amino acid substitutions F185H. Like the F185K mutation (J Biol Chem 271:7712, 1996), the F185H mutation makes the enzyme more soluble without affecting in vitro activity.
 

  
RECONSTITUTION:
 

Reconstitute lyophilized HIV-1 Integrase in 100 µL deionized H2O for 100 µg quantity. The enzyme is stable to freeze-thawing.
 

CONCENTRATION:
 
1 mg/mL in deionized H2O after reconstitution.
 
PURITY & STERILITY:
 

HIV-1 Integrase has been shown to be > 90% pure by SDS-PAGE.

STORAGE & HANDLING:
 

Store lyophilized sample at -20C or -70C prior to reconstitution. Once the integrase enzyme has been reconstituted, store at -70C.
